1. Introduction

The aim of the study was to assess the efficacy and safety of Nolvadex (Tamoxifen Citrate) in the treatment of breast cancer in postmenopausal women. This study was conducted at the Department of Oncology at the University of Heidelberg in Germany. The participants included postmenopausal women with histologically confirmed breast cancer and had been treated with tamoxifen (Tamoxi-Nolvadex). The study was carried out in a multicentre randomised, double-blinded, phase 3 study. After a mean of 12 weeks, all patients were evaluated for their response to tamoxifen treatment. A total of 100 breast cancer patients were randomised to take one of two tamoxifen tablets (Nolvadex) and Nolvadex plus tamoxifen. The response rate was defined as no adverse events reported to the patients at the end of treatment. The primary endpoint was the treatment-free survival of the participants.

2. Materials and methods

This was a phase 3, double-blinded, open-label, multicentre, randomized, single-arm study that enrolled patients with histologically confirmed breast cancer and had been treated with tamoxifen (Tamoxi-Nolvadex) for more than 12 months. The participants received a daily dose of Nolvadex (10 mg) taken once daily for five days. The dose of tamoxifen was based on body mass index (BMI), the number of participants in the study, and the number of menopausal patients (defined as a BMI ≥ 25 kg/m2 or ≥ 30 or a BMI ≥ 28 kg/m2 or >30 kg/m2). The study was conducted in compliance with the European Medicines Agency (EMA) guidance on the safety and efficacy of Tamoxifen in postmenopausal women. In the present study, the primary objective of the study was to evaluate the safety and efficacy of Nolvadex, the tamoxifen-containing dose, and the tamoxifen group.

3. Results

A total of 100 patients were randomised to take one of two doses of Nolvadex (Tamoxi-Nolvadex). The mean number of patients in the tamoxifen group was 6.1 (3.1 to 9.9) and mean treatment period was 17.6 (14.4 to 31.6) weeks, with a mean of 2.8 (0.6 to 4.9) cycles. The patients in the tamoxifen-treated group had a higher number of patients and a greater mean duration of tamoxifen therapy compared to the patients treated with the placebo (14.6 (6.6 to 20.5) vs. 3.1 (1.9 to 4.0), p<0.05). In the tamoxifen group, the mean duration of tamoxifen therapy was significantly longer than in the tamoxifen group and was more frequent in patients on Nolvadex (14.3 (5.9 to 21.7) vs. 4.8 (3.1 to 9.6), p<0.01). The mean number of patients in the tamoxifen group in the first two cycles was significantly higher than in the tamoxifen group and was greater in patients on Nolvadex (8.3 (2.6 to 19.1) vs. 4.8 (2.6 to 6.1), p<0.01). The treatment duration was significantly longer in the tamoxifen group (17.6 (13.2 to 33.8) vs. 8.9 (2.2 to 17.7), p<0.01) and was significantly more frequent in the tamoxifen group (15.1 (5.3 to 23.6) vs. 3.9 (1.6 to 7.6), p<0.01). The mean number of patients in the tamoxifen group was also significantly higher than in the tamoxifen group. The median number of patients in the tamoxifen-treated group was 7.4 (5.1 to 13.4) with a mean of 3.4 (0.6 to 4.2) cycles. The mean duration of tamoxifen therapy was significantly longer in the tamoxifen-treated group (17.7 (13.6 to 32.5) vs. 6.7 (5.4 to 8.9), p<0.01).

We’ve all been there. That’s when the real wonder drug is launched. Tamoxifen, a drug that was originally developed for breast cancer, is available on the NHS. The first real example of this kind of treatment is a very interesting study that took place in Canada.

The study took place at the University of British Columbia and looked at a patient who had previously used an aromatase inhibitor, such as Nolvadex and Femara. The results showed that the patient had been treated with the drug in Canada for about 12 months.

“We were able to say, ‘Hey, this is really the best drug for you. I think this is pretty close to where you’d be today. The first time you’ve had an aromatase inhibitor, you’ve had two years of chemo,’’ says Dr. John Stolley, professor of gynecology and immunology at the University of British Columbia.

“It really is an amazing drug, but it doesn’t really work very well. It has side effects that are very rare.’’

Tamoxifen, originally developed as an anti-estrogen, was developed to be an aromatase inhibitor. It was originally intended as a treatment for breast cancer but it wasn’t designed to be used for this type of cancer. The study was conducted at the University of British Columbia and looked at how well it worked for patients with advanced breast cancer. The results showed that the patients had been treated with the drug in Canada for about 12 months.
